研究概要

からだの主成分であるタンパク質は、その多くが糖鎖の修飾を受けて機能分子として働いています。糖鎖も含めた理解があって本来の機能を知ることができますが、ほかの機能分子に比べ、糖鎖は圧倒的に調べにくいことが課題でした。
私たちは、複数の独自糖タンパク質解析・利用技術を開発し、複合的なオミクス解析に組み込むことで、生命の理解を深め医療に貢献する研究を進めています。

研究課題

研究課題1:タンパク質翻訳後修飾解析技術開発

主要な翻訳後修飾の一つである糖鎖修飾は、タンパク質の立体構造や細胞内局在、他分子との相互作用等を変化させることで、タンパク質機能を制御することが知られています。
このようなタンパク質上糖鎖の生体機能を理解するためには、疾患や生理状態の変化に伴い、どのような糖鎖修飾機構の変化が生じ、その結果どのようなタンパク質のどの部位にどのような構造の糖鎖修飾がどの程度発現するのかを、特定糖タンパク質、細胞、組織、個体の各レベルで統合的に理解することが重要です。
産総研では、この重要課題に多角的に取り組むため、グライコプロテオミクス、グライコミクス、トランスクリプトミクス、メタボロミクス等のマルチオミクス技術の開発を進めています。また、これらの開発技術により取得したデータを糖鎖科学コミュニティで広く活用できるよう、データベース開発・公開にも積極的に取り組んでいます。

研究課題2:細胞糖鎖機能解析

細胞が正常なコントロールから外れて、がんなどの疾患状態になると、糖鎖構造が変化することが知られています。産総研では、疾患における特定のタンパク質上の糖鎖変化を疾患バイオマーカーや創薬標的として活用する研究開発を行なってきました。しかしながら、変化した糖鎖がどのような機能を持つのかは、ほとんど解明されていません。そこで、正常状態と疾患状態の個体、組織、細胞、分子における糖鎖変化を、糖鎖遺伝子、糖タンパク質、タンパク質、分解物などを対象に、多様な網羅的解析技術を用いて多角的に解析を行います。マルチオミクス解析から得られるデータと、細胞増殖、転移能、分化能、細胞死などの細胞機能解析や、がん、免疫、生殖、老化、ウイルス感染などの疾患に関連する病態表現型解析の結果を統合することにより、糖鎖の構造と機能の関係を統合的に理解することを目指しています。
